This application note describes the fully automated and in-depth characterization
of monoclonal antibody (mAb) charge variants by four-dimensional liquid
chromatography/mass spectrometry (4D-LC/MS) using the Agilent InfinityLab
2D‑LC Solution and the Agilent 6545 LC/Q-TOF system. Charge variants resolved by
cation-exchange chromatography (CEX) are collected in loops installed on a multiple
heart-cutting valve and consecutively subjected to online desalting, denaturation,
reduction, and tryptic digestion prior to LC/MS-based peptide mapping.
